Is it possible to convert metric file to Inch file?
1 Simply change the unit system (this won't change the dimensions i.e 25.4mm will be 1" )
2 Change unit system and use scale macro (ModelRescaler) to change 1mm to 1"

Once we change unit do we need to do anything to get my mm value to converted to inch in model?
If you are OK with 25.4" showing as 1" then you don't need to do anything. But if you need 25.4mm to show as 25.4" then you need to scale up the dimensions. The macro I pointed to in above post can come handy for that.

My requirement is to replace a metric stud which I added from tool box with an Inch (UN) stud.
But I could not find one in tool box so I am planning to do a save as of the metric stud and now it is done.
Now I would like to make changes in the configuration, but not able to find the design table inside configuration manager tab. Could you please help me on it.
Please check if there is a design table or not.
Also check under Tools > Options > FeatureManager and make sure design table is set to automatic and not hide.

The option which you suggested is by default set as automatic.
Then I don't think your file has design table. You can verify this by looking at the icon next to config name. If there is a yellow box then no DT and if there is a green cross then DT is there.
Insert one and then you can use that.
So how can I create or edit, Please find the below image of how my model configuration looks like.
Insert > Table > Design Table.
The Configure Feature function was used to create those configs. No need for a Design Table unless you have specific reasons to do so.
