No, symmetry is applied as a Fixture, not as part of a contact set.

What I am asking is not how to use the symmetry fixture but if I need to add the cut faces in a no penetration contact set in the presence of a symmetry fixture. I don't know if this makes sense. In the attachment I posted, I would normally only pick the front and outside/inside surfaces of the parts If I didn't have a symmetry fixture. Would I have to pick the cut surfaces as well in the presence of the this fixture?
No, you do not apply the no penetration contact definitions to the cut-faces; any contact motion on the cut-face of symmetry would by definition violate the assumptions of symmetry.

symmetry is only applied to the faces that are cut
IE where the part would be mirrored to create the full model
