Analysis with rooms at a negative depression

I am a new user trying to complete an analysis of a series of joined rooms that need to eachg need to be at a negative depression to the preceeding one. I know what pressure I require inside each room as well as the volumetric air flow (number of air changes / hour I require in each space). I have seperated each room and am trying to run each one seperately using ones rooms desired internal pressure as the input environmental pressure for the next and applying the required exit volumetric flow rate but I'm not getting the required internal pressure in that room...any ideas, suggestions? Honestly, I haven't even looked at HVAC/Fluids in over ten years...any help short of me taking time off to relearn everything would be greatly apppreciated, thanks.
