We experienced an issue when we carried out the "Defeature" command recently.
After going through all the steps of selecting components to keep and so forth, the preview window showed an empty screen although there was a solid body highlighted in the feature manager tree.
We saved the model under a new file name, open it and the strange thing was that the model appeared!
I understand that this does not solve your problem but I often get requested for models to be sent to our clients, thus I use the Defeature command in order keep our design content to a minimum when sending to a 3rd party.
We are using SW 2011 sp 2, Windows XP (x32), NVIDIA Quadro FX 1700.
What exactly are you trying to Defeature in your assembly? Are you able to Pack and Go your assembly and attached the zip file to this posting for others to have go perhaps?

Have been having issues with this tool myself. When I make a defeatured part from an assembly the part measures larger then the original part features. An edge on the original part measures 5.750. The same edge on the defeatured part measures 5.767.
