How to apply tangent relation to constrain the sketched spline.
Tangent relation between surface spline and sketched spline. Tangent symbol should display in sketch as well as the surface.

have you got 'enable spline tangency and curvature handles' checked in system options?
Yes it is enabled but not working.
Try making an intersect curve through the surface and make tangent to that.
I agree with Roland, that is my favorite way to create a tangent to face relation that is pretty robust.
Seems to be good option. I tried but not able to fix it due to irregular profile.

There you can see tangent constrain between spline middle (sketched) and surface tangent. This is what i wanted. Do you have any idea on how to do this.
it's difficult to see what you are trying to achieve. but in any instance, it appears your spline ends do not connect to any feature that would help define them, ie a tnagent edge or another sketch entity.
Assuming you're trying to connect the straight face to the round one, it would be worth defining the edges of both by either trimming them or adding split lines. Also assuming the spline doesn't need to be a 3D spline, if you insert a plane, or extrude a surface to act as a sketch plane, you could them easily add a spline between the two defined edges.
