Yes, but it requires a custom BOM based on Excel.

I only work with pre-existing .slddrw files,
and so the BOM is already on the sheet.
I wonder what/how would be the fastest/easiest way to edit the BOM,
just so that the item numbers and the coordinating balloons
have leading zeros?

Pretty sure this only applies to numerical values that happen to be in the BOM, not item numbers. I just set both those to "Show" and created a BOM. Item numbers are just 1-43 with no leading zeroes.
Yes, that's not for item numbers, but for part numbers instead.
Just double-click the "item number" cell and type your leading zero. Ignore the dire warning message that comes up. Balloons will stay linked.
