There is a bit of a bug with route lines going to holes that are on cylinders. The software tends to screw up a lot on them. I think it has to do with the fact that the software is trying o add a concentric constraint to the edge and can't find it because it isn't a circle.
What I do is place the route lines like you have them, then manually adjust them afterwards to be what I want by adding concentric constraints. Its a pain, but that is the way it works.
When selecting the face or edge that begins the route, check the direction of the arrow. That controls which direction that SEGMENT of the route moves in.
Where you select is a big factor as well... Selecting the side of the cylindrical face closest to the direction the route path is intended will make the direction of the route easy to choose.
If the direction is not the direction desired on selection, select the reverse radio button in the dialog.
