Is it possible to change this line into Construction Line so that three region has become one region for extrusion purpose?
You can either use the trim command to remove those sections or you can split the long line & make the middle section construction or make the whole line construction & then add two short lines on topadding relations to the end points or you can leave as is & select the three closed regions using contour select.

Use the Split Entities command on the Sketch tab.
If you still want to split, then right click on line in the sketch mode and select from the pop up menu

My ultimate intention is knowing split tool.
You cannot turn "part" of a line into construction...but you can divide the line into multiple lines, which will then allow you to make them construction. But for what you are doing, I do not see the need. Just trim the lines.
You can still keep the three regions and while extrusion, select all the contours and extrude . So no need to split or any trimming
Maha, just like we discussed yesterday and like Deepak showed you, you can have multiple closed sections in a sketch.
