For load time savings... Configurations or Display states un-linked from configurations?
We are having major load time issues and STP file creation issues (45 minutes)

Have you checked what features or parts are causing the slow response? You can use Assembly Visualization and AssemblyXpert. Maybe that helps already.
I still have to dive into display states and write a blog post on it. I still have no idea what they are and why I need them, so I can't answer your question yet.
display state is way faster than configs, no doubt about it.
But there are many other factors that may affect your load time, for instance: image quality read this.
As Siavash stated, display states can save time if the users are trained in using them properly. Configurations managing suppression states could work, but that opens a new can of worms with the mate management.
I am pretty sure that the reasons for your assemblies slow-down could be pin-pointed and addressed.
Solving Large Assembly slow-downs is like treating cancer. You need to find the cause of slow-down and customize the treatment.
I am doing this almost on a full-time basis now, and I still discover every day something new.
Example: Today I found a large assembly that takes 15 minute to open. Even worse, if you add a component, or add a mate, or just look at it the wrong way would trigger a 10 minute rebuild. The users practically work 3 minutes every hour. The rest is spent waiting for the machine to rebuild the assembly (especially to re-generate the graphic triangles).
This is what I found that is actually new to me. One of the parts is an imported file with lots of topological errors. There are so many, that solving them is impossible. If the Image Quality setting for this part is bigger than 20%, the computation of the tessellation (graphic-triangles) takes upward of 10 minutes.
If I drop the Image Quality setting to about 10%, the tessellation takes 5 seconds. I did that, and the users can open now the assembly in 18 second and rebuild it in 5 second.
