have problem with fatigue study principles and how i should apply it for true results

I have the attached model, diaphragm coupling, and I applied constant torque to one hub and fixtures such that make parallel miss alignment in model. As you know miss alignment makes variable stress in every point of model in a cycle so fatigue study is more important. I have static results and now I want to simulate it with fatigue analysis. How I can apply the fatigue study and get true results agree with this parallel miss alignment? If I should use variable amplitude fatigue study what load history curve I should apply to it?
